I was away for a long time so I wonder if we can use markdown syntax in posts? I see that I can use those buttons in the editor but I feel more comfortable if I don't need to move my fingers away from the keyboard.

No can not us markdown in post,MyBB use BBcode and many of tags have been rewritten bye us to get eg code syntax highlighting in posts.
If want to try some in post can look at Markdown to BBcode,but as mention we have rewritten many of the tags.
