Everything before mainloop() happens until you close the Tkinter window. The rest, the counting part, doesn't happen until you exit the GUI.

Grid_Cell_id is a file pointer. You don't ever read anything from the file. Reading a file is in every beginner tutorial.

You first have to tell the function, on_click, which button was clicked. Then you can use the config option to get the current background color and change it accordingly.
